Endoplasmic reticulum, oleosins, and oils in seeds and tapetum cells.

چکیده
Diverse organisms contain neutral lipids in subcellular particles for food reserves and other purposes. These lipid particles are present in seeds, flowers, pollen and fruit of higher plants, the vegetative and reproductive organs of primitive plants, algae, fungi, nematodes, mammalian glands and brown adipose tissue of mammals, and bacteria. Of all these lipid particles, the oil bodies (OBs) in seeds are the most prominent and best studied. Seeds of most plant species store oils (triacylglycerols [TAGs]) as a food reserve for germination and postgerminative growth. TAGs are present in small subcellular spherical OBs of approximately 1 mm in diameter. Each OB has a matrix of TAGs surrounded by a layer of phospholipids (PLs) and structural proteins termed oleosins. The small size of OBs provides a large surface area per unit TAG, which would facilitate lipase binding and lipolysis during germination. OBs inside the cells of mature seeds or in isolated preparations are remarkably stable and do not aggregate or coalesce. This stability is in contrast to the instability of artificial liposomes made from amphipathic and neutral lipids; the liposomes gradually coalesce after formation. Seed OBs are stable because their surface is shielded by a layer of oleosins. In maturing seeds, TAGs, PLs, and oleosins are synthesized in the endoplasmic reticulum (ER), from which budding OBs are released. Research on seed OBs and oleosins has been reviewed byHuang (1992, and an earlierUpdate article in 1996), Napier et al. (1996), Galili et al. (1998), Frandsen et al. (2001), and Murphy (2001). This Update provides brief reviews of earlier work and emphasizes recent major discoveries. Detailed information and references to earlier work can be found in the previous reviews.

متن کاملTapetosomes in Brassica tapetum accumulate endoplasmic reticulum-derived flavonoids and alkanes for delivery to the pollen surface.
Tapetosomes are abundant organelles in tapetum cells during the active stage of pollen maturation in Brassicaceae species. They possess endoplasmic reticulum (ER)-derived vesicles and oleosin-coated lipid droplets, but their overall composition and function have not been established. In situ localization analyses of developing Brassica napus anthers revealed flavonoids present exclusively in ta...
متن کاملA STUDY ON ULTRASTRUCTURAL ASPECTS OF SEED DORMANCY IN WILD OATS
The structural differences between dormant and nondormant seeds of wild oats (Avena ludoviciana Durieu) were studied by light and transmission electron microscopy. Radial elongation of scutellum epithelial cells were observed in nondormant seeds after imbibition, compared with dormant seeds in which cells were completely compressed. Ultrastructural differences for example, dilution of cytopla...
متن کاملGating Behavior of Endoplasmic Reticulum Potassium Channels of Rat Hepatocytes in Diabetes
Background: Defects in endoplasmic reticulum homeostasis are common occurrences in different diseases, such as diabetes, in which the function of endoplasmic reticulum is disrupted. It is now well established that ion channels of endoplasmic reticulum membrane have a critical role in endoplasmic reticulum luminal homeostasis. Our previous studies showed the presence of an ATP-sensitive cationic...
متن کاملBioinformatics Reveal Five Lineages of Oleosins and the Mechanism of Lineage Evolution Related to Structure/Function from Green Algae to Seed Plants.
Plant cells contain subcellular lipid droplets with a triacylglycerol matrix enclosed by a layer of phospholipids and the small structural protein oleosin. Oleosins possess a conserved central hydrophobic hairpin of approximately 72 residues penetrating into the lipid droplet matrix and amphipathic amino- and carboxyl (C)-terminal peptides lying on the phospholipid surface. Bioinformatics of 1,...
